Three poly(amidoamine)s with pendant primary amines (polymers 1a-1c) were synthesized by Michael polyaddition of N-Boc-protected diamine to N, N-methylene-bis(acrylamide), followed by the removal of N-Boc-protective groups under acidic conditions. The poly(amidoamine)s with primary and tertiary amines possess a high buffer capacity between the pH 5 to 7, which facilitates polymer/DNA complexes to escape from endosomes according to the “proton sponge hypothesis.” In vitro cytotoxicity of poly(amidoamine)s was dependent on the side-chain structure of the polymer, namely, the cytotoxicity increased with increasing the length of the side chains. Polymers 1a-1c could condense plasmid DNA via electrostatic interaction to form nanosized (100-500 nm) polyelectrolyte complexes with positive surface charge. In vitro transfection results indicated that the transfection efficiency of polymer 1b under optimized conditions was comparable to that of commercial branched PEI 25000.
